Effective Inhibition of Glomerulosclerosis by Adenoviral Vector Expressing Human IL-10

|
Focal segmental glomerulosclerosis (FSGS) causes severe renal injury that exhibits tubular atrophy, glomerular sclerosis and increment of proteinuria, associated with progressive functional loss of the kidneys. Although the mechanism causing renal failure in FSGS is not fully defined, FSGS shows interstitial inflammation by immune stimulation cells including T cells and B cells. This study aimed to study the utility of IL-10, a cytokine with potent anti-inflammatory activity, in the treatment of FSGS in FGS/Kist mice in which FSGS occurs naturally with aging. Groups of six mice received recombinant adenoviruses encoding human IL-10 at 6 weeks and were investigated for glomerular index and proteinuria. The group treated with IL-10 was shown to have significant suppression of glomerular sclerotic index (male: 1.3%, female: 0.25%) at 10 weeks when compared to LacZ (male: 54%, female: 10%) or sham controls (male: 69%, female: 12%). We also observed reduced proteinuria in FGS/Kist mice transduced with recombinant adenoviruses harboring human IL-10. These results demonstrate that IL-10 prevents FSGS in FGS/Kist mice in an effective manner and gene therapy using IL-10 may be used for the treatment of renal failure.
